(Examinateur)<\/li>\n<\/ul>\n<h5>Summary:<\/h5>\n<p>Alors que 50 million de personnes souffrent de traumatismes cr\u00e2niens chaque ann\u00e9e dans le monde entier, le choc (stress m\u00e9canique externe), engendr\u00e9 \u00e0 cette occasion sur le r\u00e9seau neuronal, peut amener \u00e0 l\u2019apparition de l\u00e9sions c\u00e9r\u00e9brales traumatiques. A moyen et long termes, ce processus peut entra\u00eener une neuro-inflammation et la manifestation de pathologies telles que la maladie de Parkinson ou d&rsquo;Alzheimer. Ce travail de doctorat a pour objectif de d\u00e9velopper un microsyst\u00e8me \u00e9tirable pour l\u2019\u00e9tude \u00e9lectro-m\u00e9canique de r\u00e9seaux de neurones in vitro. Un tel microsyst\u00e8me doit int\u00e9grer des \u00e9lectrodes isolantes, m\u00e9caniquement robustes sur le support \u00e9tirable. La premi\u00e8re partie de ce travail a consist\u00e9 en l\u2019\u00e9tude de la m\u00e9tallisation directe sur PDMS par masquage physique. Ensuite, une couche mince de la r\u00e9sine photosensible \u2018SU-8\u2019 a \u00e9t\u00e9 intercal\u00e9e entre le PDMS et les m\u00e9taux permettant ainsi de rigidifier les lignes m\u00e9talliques et d\u00e9placer la contrainte m\u00e9canique vers le support de PDMS. Afin d\u2019\u00e9viter toute fissure sous contrainte m\u00e9canique, l\u2019architecture et l\u2019orientation des \u00e9lectrodes a \u00e9t\u00e9 \u00e9tudi\u00e9e et valid\u00e9e par un mod\u00e8le physique. Dans un deuxi\u00e8me temps, un microsyst\u00e8me de PDMS\/SU-8\/Cr-Au\/Paryl\u00e8ne a ensuite \u00e9t\u00e9 fabriqu\u00e9 par photolithographie via une technologie planaire et est compatible avec le dispositif commercialis\u00e9 par MultiChannel System\u00a9 qui permet de visualiser et d\u2019enregistrer les influx nerveux d\u2019un r\u00e9seau neuronal mature. Pour la fabrication du microsyst\u00e8me \u00e9tirable, un proc\u00e9d\u00e9 de microfabrication complet a \u00e9t\u00e9 d\u00e9velopp\u00e9 pour 3 \u00e9tapes de photolithographie sur PDMS. La fonctionnalit\u00e9 de ce microsyst\u00e8me a \u00e9t\u00e9 valid\u00e9e par la visualisation de r\u00e9ponses \u00e9lectriques d\u2019un r\u00e9seau neuronal au bout de 12 jours in vitro (DIV). Ce microsyst\u00e8me pr\u00e9sente des \u00e9lectrodes avec un ratio signal\/bruit comparable aux MultiElectrode Array (MEA) commerciaux<\/p>\n<h5>Abstract:<\/h5>\n<p>50 million people suffer from head trauma each year worldwide. The shock (external mechanical stress) acting on neural networks insided the brain can lead to the appearance of traumatic brain injuries (TBI). In the short term and long term, this process can lead to neuroinflammation and the manifestation of pathologies such as Parkinson\u2019s disease or Alzheimer\u2019s disease. This thesis aims to develop a stretchable microsystem for the electromechanical study of neural networks in vitro. Such a microsystem must integrate insulated electrodes which are mechanically robust on the stretchable support. The first part of this work consisted in studying the direct metallization on PDMS using physical masking. Next, a patterned photoresist (SU-8) thin film was then used between the PDMS and the metals. The stiff SU-8 shields the metallization from strain\u2014which now occurs in the adjacent PDMS. In order to avoid cracks under mechanical stress, the architecture and orientation of the electrodes has been studied and validated by a physical model. The second part was the fabrication of a PDMS\/SU-8\/Cr-Au\/Parylene stretchable microsystem by photolithography using a planar process developed during the thesis work. The microsystems is compatible with MultiChannel System\u00a9 commercial device which allows the visualization and recording the nerve impulses of a mature neural network. A microfabrication process has been developed with 3 photolithography steps on a PDMS substrate. The functionality of the microsystem has been validated by the visualization of electrical responses of neural network at 12 days in vitro (DIV).
