th\u00e8se)<\/li>\n<\/ul>\n<h5>Summary:<\/h5>\n<p>Malgr\u00e9 la disponibilit\u00e9 de diff\u00e9rents antibiotiques, les infections bact\u00e9riennes demeurent l\u2019une des principales causes d\u2019hospitalisation et de mortalit\u00e9. L\u2019\u00e9chec clinique du traitement antibiotique est d\u00fb \u00e0 une faible p\u00e9n\u00e9tration g\u00e9n\u00e9rale des antibiotiques dans les sites d\u2019infection bact\u00e9rienne ainsi qu\u2019au d\u00e9veloppement de pathog\u00e8nes r\u00e9sistants aux antibiotiques. La mise au point de strat\u00e9gies antibact\u00e9riennes alternatives moins soumises aux forces s\u00e9lectives \u00e0 l\u2019origine de l\u2019apparition d\u2019une r\u00e9sistance acquise aux antibiotiques est devenue un besoin urgent. Les temp\u00e9ratures sup\u00e9rieures \u00e0 70 \u00b0C provoquent g\u00e9n\u00e9ralement une destruction instantan\u00e9e des bact\u00e9ries. L\u2019ablation bact\u00e9rienne par la chaleur est par cons\u00e9quent une strat\u00e9gie int\u00e9ressante \u00e0 poursuivre. Dans cette th\u00e8se, l\u2019ablation photothermique d\u2019agents pathog\u00e8nes est \u00e9tudi\u00e9e. Le d\u00e9veloppement d\u2019un patch cutan\u00e9 photothermique flexible \u00e0 base d\u2019un film mince de poly (imide), Kapton, modifi\u00e9 avec de l\u2019oxyde de graph\u00e8ne r\u00e9duit (rGO) a permis un traitement rapide et tr\u00e8s efficace des infections de plaies sous-cutan\u00e9es par irradiation dans le proche infrarouge. L\u2019isolement magn\u00e9tique et sp\u00e9cifique dans le s\u00e9rum de E. coli responsable d\u2019infection des voies urinaires (UTIs) \u00e9tait possible \u00e0 l\u2019aide de nanocomposites rGO magn\u00e9tiques avec des anticorps anti-fimbrial E.coli int\u00e9gr\u00e9s. Un \u00ab\u00a0nanotransporteur\u00a0\u00bb magn\u00e9tique optimis\u00e9 a atteint une efficacit\u00e9 de capture de 99,9%, m\u00eame \u00e0 des concentrations de 1\u00d7101 cfu mL-1 de E. coli en 30 min. L\u2019irradiation du nanocomposite charg\u00e9 de E. coli avec un laser proche de l\u2019infrarouge a entra\u00een\u00e9 l\u2019ablation totale des agents pathog\u00e8nes \u00e9limin\u00e9s.<\/p>\n<p>Parall\u00e8lement, l\u2019administration de m\u00e9dicaments transdermiques \u00e0 base de chaleur a \u00e9t\u00e9 envisag\u00e9e pour l\u2019administration d\u2019antibiotiques et d\u2019autres m\u00e9dicaments tels que la metformine. Des hydrogels de graph\u00e8ne (GO)\/metformine et d\u2019hydrogel de graph\u00e8ne r\u00e9duit enrichi en acide carboxylique (rGO-COOH)\/metformine ont \u00e9t\u00e9 notamment fabriqu\u00e9s, leur profil de chauffage ainsi que la perm\u00e9abilit\u00e9 \u00e0 travers la peau de la metformine ont \u00e9t\u00e9 d\u00e9termin\u00e9s.<\/p>\n<h5>Abstract:<\/h5>\n<p>Despite the availability of different antibiotics, bacterial infections are still one of the leading causes of hospitalization and mortality. The clinical failure of antibiotic treatment is due to a general poor antibiotic penetration to bacterial infection sites as well as the development of antibiotic resistant pathogens. The development of alternative antibacterial strategies that would be less subject to the selective forces that drive the emergence of acquired antibiotic resistance has become an urgent need. Temperatures higher than 70\u00b0C cause generally an instant killing of bacteria. Heat based bacterial ablation is consequently an interesting strategy to pursue. In this thesis, photothermal ablation of pathogens is investigated. The development of a flexible photothermal skin patch based on a poly(imide) thin film, Kapton, modified with reduced graphene oxide (rGO) allowed for a rapid and highly efficient treatment of subcutaneous wound infections via near-infrared irradiation. Magnetic and specific isolation of E. coli associated with urinary tract infection (UTIs) from serum was possible using magnetic rGO nanocomposites with integrated anti-fimbrial E.coli antibodies. An optimized magnetic nanocarrier achieved a 99.9 % capture efficiency even at E. coli concentrations of 1\u00d7101 cfu mL-1 in 30 min. Irradiation of the E. coli loaded nanocomposites with a near-infrared laser resulted in a total ablation of the eliminated pathogens.<\/p>\n<p>In parallel, heat-based transdermal drug delivery has been considered for the delivery of antibiotics and other drugs such as metformin.
