Gr\u00e2ce \u00e0 l\u2019application de diff\u00e8rent techniques (Essential Dynamics, Steered MD, \u2026) on a caract\u00e9ris\u00e9 les stades pr\u00e9coces de l\u2019\u00e9volution de cette l\u00e9sion d\u2019ADN dans les deux \u00e9l\u00e9ments.<\/p>\n<h4><strong>Abstract:<\/strong><\/h4>\n<p>This thesis is dedicated to a combined theoretical and experimental investigation of the structure and dynamics of two common types of defects occurring in the DNA molecule, after chemical or radiation damage: base mismatches and strand breaks.<\/p>\n<p>We used single-molecule force spectroscopy performed by optical tweezers accompanied by Molecular Dynamics (MD) all-atom simulations, to characterize mismatches in short DNA hairpins. We demonstrate that it is possible to use SMFS<\/p>\n<p>Subsequently, we designed structural models for the DNA strand-break defects, in the two key constitutive elements of the chromatin: the DNA linker and the nucleosome.
