Normalement, les \u00e9tudes de r\u00e9paration sont trait\u00e9s par des exp\u00e9riences sur des brins d\u2019ADN uniques et isol\u00e9s. Par contre, tous les processus de r\u00e9paration cellulaire <em>in vivo<\/em> se font dans le contexte de la chromatine, la structure densement repli\u00e9e que l\u2019ADN assume dans le noyau, en s\u2019enroulant sur des c\u0153urs de prot\u00e9ines (les histones), et cr\u00e9ant ainsi une \u00e9troite s\u00e9quence de \u00ab\u00a0perles sur fil\u00a0\u00bb. Ces perles sont les <strong>nucl\u00e9osomes<\/strong>, que l\u2019on peut consid\u00e9rer comme les briques \u00e9l\u00e9mentaires de la chromatine et des chromosomes. De mani\u00e8re originale, le projet DYPROSOME cible ainsi l\u2019\u00e9tude des m\u00e9canismes de r\u00e9paration par BER dans les nucl\u00e9osomes.<\/p>\n<p>Les partenaires du IAB Grenoble et LBMC Lyon, parmi les sp\u00e9cialistes majeures dans leurs domaines, assemblent des \u00e9chantillons de nucl\u00e9osomes reconstruits en laboratoire, avec des d\u00e9fauts ins\u00e9r\u00e9s dans la s\u00e9quence d\u2019ADN, imitant le r\u00e9sultat de l\u2019endommagement, tels que typiquement apport\u00e9s \u00e0 la cellule par des agents pathog\u00e8nes chimiques, des processus d\u2019oxydation et vieillissement cellulaire, ou l\u2019interaction avec les rayonnements ionisants. Ces nucl\u00e9osomes \u00ab\u00a0endommag\u00e9s\u00a0\u00bb sont mis en solution avec les enzymes de la BER d\u00e9put\u00e9s \u00e0 la reconnaissance des d\u00e9fauts, pour \u00e9tudier leur structure en interaction avec l\u2019ADN endommag\u00e9. Les d\u00e9tails de la structure mol\u00e9culaire sont r\u00e9v\u00e9l\u00e9s par la cryo-microscopie atomique.<\/p>\n<p>En parall\u00e8le, les partenaires de l\u2019IEMN et UGSF, tous les deux bas\u00e9s \u00e0 Lille, r\u00e9alisent des simulations \u00e0 l\u2019ordinateur par des m\u00e9thodes de <em>docking<\/em> (amarrage) mol\u00e9culaire et de dynamique mol\u00e9culaire, visant \u00e0 reconstruire les d\u00e9tails structurels et dynamiques \u00e0 l\u2019\u00e9chelle atomique des interactions enzymes\/ADN.
